Avrainvillamide is the monomer of the stephacidin B dimer. Avrainvillamide is active against the proto-oncogene, mutated NPM1, commonly present in patients with acute myeloid lukemia. Avrainvillamide alkylates Cys275 of mutated NPM1, inhibits nuclear export of Crm1 cargo proteins. Aavrainvillamide also displaces Thr199-phosphorylated NPM1 from duplicated centrosomes, leads to an accumulation of supernumerary centrosomes and inhibits dephosphorylation of Thr199-phosphorylated NPM1 by protein phosphatase 1.
