    Brefeldin A is a lactone antiviral involved in protein transport inhibition and tumor apoptosis.  It was first isolated from the fungus Penicillium decumbens in 1958 by VL Singleton who named it Decumbin.  The chemical structure of the compound was elucidated in 1971 by H.P. Siggs.

    Mechanism of Action

    Brefeldin A inhibits protein transport by preventing accumulation of COPI proteins on the Golgi apparatus which facilitate vesicle fusion from the endoplasmic reticulum (ER). By preventing Golgi-COPI protein function, vesicles from the ER cannot properly fuse with the Golgi to be secreted to destined locations.

    Cancer Applications Brefeldin A has demonstrated anti-cancer properties by inducing apoptosis in leukemia and colon carcinoma cells.
