    De-Plasma™ II

    De-Plasma™ II is a safe, potent reagent which can be used to prevent or eliminate Mycoplasma contamination.  It consists of a proprietary mixture of five antimicrobial compounds with three separate anti-Mycoplasma targets.  It is non-toxic to most cell lines with an ED50 10X larger than the treatment concentration. 

    Mycoplasma belong to a unique genus of bacteria that lack cell walls.  Their small size and ubiquitous distribution account for their ability to go undetected in seemingly sterile cell cultures, but ultimately cause alterations in cell physiology and metabolism, leading to inaccurate experimental conclusions.

    De-Plasma™ II is an antibiotic cocktail comprised of one macrolide, one quinolone, and one tetracycline compound, each of these groups have shown they are highly active against Mycoplasmas.  This mixture offers higher potency and is effective against all known Mycoplasma species and related non cell-walled prokaryotes.

    Mechanism of Action The mechanism of action for each of the antibiotics in De-Plasma II are different. Tetracyclines and macrolides block protein synthesis by interfering with translation at the prokaryotic ribosome. Quinolones inhibit bacterial DNA gyrase and topoisomerase IV, blocking DNA replication. Eukaryotic cells do not contain DNA gyrase or topoisomerase IV and are therefore unaffected by De-Plasma II. 
    Microbiology Applications De-Plasma II is a safe, effective anti-Mycoplasma reagent to avoid or eliminate Mycoplasma contamination. Use at 5 μg/ml for primary cells in culture and 2.5 μg/ml for prophylactic use.  
    Cell Culture Applications De-Plasma was used to routinely test carcinoma cell lines (HNSCC cell line) including SNU1076, FADU, HSC-2 and HSC-4 (Bdarny M et al, 2018). De-Plasma was also used to authenticate the human papilloma virus negative cells (Elkabets et al, 2015).
    Protocols De-Plasma II can be added directly to the culture medium or to the flask containing the cells. We recommend preparing a 1000x stock solution (5 mg/ml).  To prepare, add 1 ml H20 to the 25 mg vial of De-Plasma II, shake, and transfer to a separate tube and then add another 4 ml H20 to make 5 ml of stock solution.  The solution can be stored at 4°C for up to 3 months.

    Working concentration: 5 μg/ml

    Decontamination period: 4 passages, 2-3 week duration each passage

    Concentration for maintenance of Mycoplasma-free culture: 2.5 μg/ml
